Miss Maudie usually makes small cakes for the children, so her giving Jem a slice of the big cake symbolises that he is no longer a child and is becoming a man. This is shown in the quote "We understood when she cut from the big cake and gave a slice to Jem".

The larger slices of cake were previously only given to adults, so by giving him one Miss Maudie recognises that Jem is becoming a man.

Ms Maudie giving Jem a big piece of cake means that she is recognising that he is becoming an adult. I agree as Jem (and Scout and Dill) previously would smaller pieces of cake because they were children.
